Name of the project: Signaling and regulation of stem cells in the adult brain

Goals and objectives

Research directions: Research in stem cells and neurogenesis in the adult brain; investigation of impact of radiation on neurogenesis and behavior; development of new methods for detecting neurogenesis and 3D analysis; reaction of new genetically encoded sensors for redox and calcium imaging

Project objective: Development of new approaches to research of processes of division, differentiation and signaling of brain stem cells and their offspring and usage of these approaches for determining the mechanisms of action of factors affecting neurogenesis.


The practical value of the study

  • We have developed new histochemical methods for detecting proliferating cells in integral brain samples and mathematical methods of automatic quantitative and qualitative analysis of 3D images.
  • Our researchers have developed a method of multi-marker labeling of dividing cells in biological tissues and determining length of the S-phase of the cell cycle and entry of cells into recurrent cell divisions.
  • We have developed and experimentally tested a method for estimating probability of symmetric and asymmetric divisions in stem cells of the brain.
  • We have developed and experimentally tested new calcium and redox indicators possessing unique sensitivity and dynamics. 
  • We have determined cell targets for influence of radiation on neurogenesis in the adult brain and investigated ability of neurogenic niches to spontaneously recover form radiation impact.
  • Our researchers have studied effects of radiation on memory and behavior of mice.

Implemented results of research:

  • Methods have been developed for staining    single-piece samples of brain cells, as well as methods of analysis of 3D images.
  • The Laboratory has obtained patents for inventions: patent No. 2563679 «A method of immunohistological staining of whole mount samples of biological tissues (options)» and patent No. 2620559 «A method of staining of whole mount samples of biological tissues and organs using click histochemistry (options)». The inventions belong to the field of experimental biology and medicine where it is necessary to prepare whole samples of biological tissues and organs for optical projection, block-face and serial two-photon tomography, confocal, multiphoton and light sheet microscopy to plot 3D maps pf histological localization of cell populations, tagged with an ethinyl marker or antibodies. These inventions can be used for pre-clinical tests of pharmacological agents and evaluation of physiological impact on the organism, as well as for working with biopsy materials for diagnostic and scientific purposes.

Education and career development:

  • 7 masters degree theses and 8 bachelors degree theses have been defended.
  • We have conducted two research and practice schools in immunohistochemistry with participation of members of the academic staff of the Laboratory.

Other results: Members of the academic staff of the Laboratory have actively participated in research activities and publicized works of the Laboratory in the media.
